Electron microscopic studies on the cells of the several organs of the rats after Yung-Sa administration

Abstract
To investigate the toxic effect of Yung-Sa (Hg compound) on several organs of the rats, electron microscopic studies were carried out on the epithelial -cells of the small intestine, hepatic cells and the renal tubular cells of the rats which are fed Yung-Sa (0. 1 mg/gm body weight/day) with ordinal rats meal for three days and five and six weeks, and results obtained were as follows. :
1. In epithelial cells of the small intestine, no significant changes in the fine morphology of the cell were noted except slight increase of the granular endoplasmic reticulum and dilation of the Golgi cisternae at three days after Yun Sa administration, but showed the decrease of the granular endoplasmic reticulum, swelling -of the mitochondria, and decrease of the, Golgi area at five .and six weeks -after Yung-Sa administration. Few Hg-particles were noted ill the cytoplasm and cell organelles of the intestinal epithelium at three days after Yung-Sa administration, and were increased and tended to concentrate in the membrane-limited structures, such as vesicles, vacuoles, and dense bodies at five and six weeks after administration of Yung-Sa.
2. In the hepatic cells decrease of nuclear materials considerable swelling of the mitochondria with increased number of the dense granules, and distortion and decrease of the Golgi area were noted at three days after Yung-Sa administration. These changes were enhanced, and histologically, various degree of! degenerative or necrotic changes in the hepatic cells were demonstrated at five and six weeks after Yung-Sa administration. Hg--particles were present free in the cytoplasm, and were concentrated in the fat droplets and dense bodies at three days of Yung Sa administration and these were significantly increased in-the hepatic cells at five and six weeks after Yung-Sa administration..
3. In the, kidney the changes were slight in the epithelial cells of the distal tubules and the collecting ducts, but were significant in the epithelial cells oproximal tubules in which¢¥ mitochondria were swollen and contained few or n dense granules at three days after Yung-Sa administration. These change were severed at five and six weeks after Yung-Sa administration. Hg-particles were noted in the cytoplasmic, metrix dense bodies and in mitochondria in almost all cells of the proximal, distal and collecting tubules at three days after YungSa administration, and tended to concentrate in the dense bodies at five and six weeks after Yung-Sa administration.
From these results, it is evident that the most toxic effects of Yung-Sa are on the mitochondria of the cells of all organs, especially of the liver and proximal tubules, in which the concentration of Yung-Sa is high.
